Morphometric and Optical Density Alterations of Red Blood Cells under PC-1 Tumor Progression and Upconversion Nanoparticle-Mediated Laser Exposure

Abstract
This work investigates the morphometric (area, diameter, thickness) and optical density (OD) alterations of rat red blood cells (RBCs) under PC-1 tumor progression, upconversion nanoparticle (UCNP) administration, and laser exposure. The optical density was calculated via light transmission measurements.
Tumor development (day 21) induced a notable decrease in RBC geometric parameters alongside an increase in OD, which is associated with cell shrinkage and denser hemoglobin packing. The injection of UCNPs alone exhibited no significant effect on RBC morphology. Furthermore, no significant deviations in optical or morphometric indices were observed in the group treated with UCNPs combined with laser irradiation. The findings highlight the diagnostic potential of RBC optical and geometric profiling during theranostic procedures.
