The effect of selecting the measuring volume under structured illumination using LED

Abstract
The thickness of biological samples often exceeds the depth of field of the microscope objective, which is why the image is affected not only by the planes that are in the focus of the objective, but also by the entire remaining thickness of the sample being measured, which is out of focus. The objective of this work was to develop a scanning laser microscope for measuring the longitudinal resolution of laser and white light in various media. The use of dynamic spatial modulation makes it possible to obtain an axial resolution of 4 μm when using laser radiation and 2 μm with a numerical aperture of the objective of 0.2.
