Single-molecule study of SMC-like protein RecN-DNA binding using optical tweezers

Abstract

The SOS response is a cascade of reactions activated in bacterial cells when DNA is damaged. Such damage can be caused, in particular, by ultraviolet radiation or the action of antibacterial drugs. Triggering the SOS response can ensure both precise repair of damaged DNA regions and bacterial adaptation to stress factors through enhanced mutagenesis. This is why the SOS response plays a key role in the development of bacterial resistance to antibiotics—one of the most pressing problems in modern medicine. This work examines the DNA-binding properties of SOS response proteins at the single-molecule level using optical tweezers.
