The semi-empirical approach to data analysis in laser diffractometry of wet blood smears

Abstract

Laser diffractometry is an indirect method of measuring the parameters of an ensemble of erythrocytes of the sample under study. In this paper, we consider the study of erythrocytes in a wet blood smear using laser diffractometry. We have already discussed the methodology of preparing a wet blood smear [1] and the required technical specifications of the experimental setup, as well as a number of algorithms for processing diffraction patterns, in particular, [2]. At the moment, our attention is focused on the experimental verification of the developed algorithms on real blood samples from healthy donors and patients' blood. The verification is as follows. We examine venous blood samples in two ways: by laser diffractometry using the diffraction pattern processing algorithms developed by us and wet smear microscopy. Taking into account the specifics of each method, we correlate the values of the average cell radius and the width of the distribution of erythrocytes by size and shape (DESS) obtained using each method. These values were chosen as the most convenient for measurement and analysis. Since other parameters of blood cells also affect the type of diffraction pattern, such verification is highly important for the further development of the laser diffractometry method.
25 venous blood smears from 8 donors were examined by us, and algorithm [2] was applied to find the above parameters of erythrocytes. It was found that the theoretically and experimentally calculated values of the DESS parameter differ, but it is possible to obtain empirically a dependence between these values. For the studied blood samples, the error in determining the DESS parameter is no more than 25%. Work in this direction continues.
