Photodynamic therapy of tumors in mice with colorectal cancer

Abstract
Balb/c mice were inoculated intramuscularly with 20 μl of CT-26 colorectal cancer cell suspension, and when the tumor volume reached 500 mm3, phodynamic therapy (PDT) was performed. Indocyanine green (IG) was used as a photosensitizer, which was diluted in polyethylene glycol at a ratio of 1:100 and administered to mice intravenously at a dose of 2 mg/kg. Doppler study of tumors was performed in mice before therapy, and after IG administration, its biodistribution was studied in dynamics using Fluor i In Vivo fluorescence imaging system (NeoScience, South Korea). One hour after IG injections, the tumor was irradiated percutaneously with 808 nm diode infrared laser LS-2-N-808-10000 (Russia) at a power density of 2.3 W/cm2 for 10 min. The temperature of local tumor heating was measured by IRYSYS 4010 thermal imager (UK). Animal withdrawal and sampling of tumor tissues for histological study were performed 72 hours after therapy. During PDT therapy the temperature rise of local tumor heating up to 40±5ºС was noted, necrotic changes in tumor tissue were observed after the therapy.
