Laser Doppler flowmetry to assess the function of the skin microvasculature in white outbred rats with diet-induced obesity

Abstract
Obesity is a widespread multifactorial disease characterized by subclinical metabolic inflammation causing the development of endothelial dysfunction (ED), which is the pathogenetic basis of cardiovascular diseases (CVD). ED is characterized by functional changes in the vascular wall, which can be diagnosed using the non-invasive method of laser Doppler flowmetry (LDF), examining the function of the endothelium of the microvasculature of the skin.
The purpose of the work is to use the LDF method to evaluate the effect of nutritional obesity and subclinical metabolic inflammation on the function of the microvasculature of the skin of the distal hind limb in white outbred rats.
The studies were performed on 20 white rats, divided into groups: 1) control (intact animals); 2) experimental group - animals with the diet-induced obesity («cafeteria diet»). Using the method of LDF on the skin of the paw, the perfusion coefficient (М) was measured. Wavelet analysis was used to determine the amplitudes normalized by standard deviation of endothelial, neurogenic, myogenic, respiratory, pulse oscillations. The levels of C-reactive protein (CRP), monocyte chemoattractant protein-1 (MCP-1), and vascular endothelial growth factor (VEGF) were determined in the blood serum.
Has been shown, that diet-induced obesity to induce subclinical metabolic inflammation, contributing to the development of ED, which is involved in both the early and late stages of vascular remodeling. The non-invasive LDF method makes it possible to diagnose ED at the preclinical stage of CVD.
