Multimodal Optical Imaging-Guide Nanotheranostics

Abstract
This presentation integrates advanced imaging techniques and therapeutic strategies, derived from three pivotal studies. The first study investigates the use of spectral-domain optical coherence tomography (SD-OCT) combined with gold nanorods (GNRs) to achieve high-resolution 2D/3D imaging of live mouse embryos. The enhanced OCT signal and penetration depth, confirmed through inductively coupled plasma mass spectrometry, facilitated improved structural visualization of embryonic organs, presenting a novel methodology for monitoring organ development and identifying congenital abnormalities. The second study introduces a multimodal imaging strategy incorporating photoacoustic (PA) imaging, OCT angiography (OCTA), and laser speckle (LS) imaging to monitor tumor angiogenesis and evaluate nanotherapeutic interventions. This approach provides detailed macroscopic and microscopic insights into tumor microvasculature and hemodynamics, thereby advancing the understanding of the therapeutic efficacy of nanotheranostics. Finally, the third study combines NIR-II PA and OCTA molecular imaging with photothermal therapy (PTT) for bladder cancer (BC) treatment. By utilizing a NIR-II hyaluronic acid-IR-1048 photothermal agent, this strategy enables dynamic monitoring of BC microvascular changes, ultimately facilitating effective minimally invasive PTT, and establishing a robust foundation for precision therapies targeting urinary system tumors. Collectively, these studies highlight the potential of multimodal imaging technologies to advance research in embryonic development, oncological treatments, and therapeutic interventions.
