Labeling and Tracking of Mesenchymal Stem Cells using Photoconvertible Microcapsules

Abstract
Сell behavior and migration (especially hMSCs) are focal points of research in the biomedical field. However, the safety of their use remains controversial owing to limited data on changes that occur in hMSCs in the long term. Fluorescent photoconvertible proteins are traditionally used as labels to study single-cell interactions, migration processes, and formation of pure lines. However, their use requires genetic modification of the primary culture, which casts doubt on the possibility of exploring cell clones in personalized medicine Our approach based on application of capsules which loaded with Rhodamine B and after thermal treating possess fluorescent photoconversion properties. Photoconvertible capsules were photostable inside the cell for a minimum of 7 days, demonstrated low cytotoxicity for hMSCs, and did not affect the immunophenotype. The developed approach provided an opportunity for cell tracking for 4days, including tracing the destiny of daughter cells without the need for additional labeling.
