The development of methodology and application of Raman spectroscopy for the detection, identification and analysis of the functional state and metabolism of microorganisms are topical tasks due to an ability of a number of microorganisms to cause diseases of plants, humans and animals; an ability to contaminate a food and technical liquids, etc.; an industrial importance and use in biotechnologies; their role in forming of ecological communities.
On the examples of microscopic algae (cyanobacteria Synechocystis sp. PCC6803 and its mutants), microbes (such as fungi of the genus Aspergillus, E. coli, etc.) amoeba the use of Raman spectroscopy is demonstrated for the detection of microorganisms and the study of the features of their structure, metabolism, and functional state under various conditions (including their interaction with environment and external effects).
Raman spectroscopy of microorganisms is often complicated by the low intensity of Raman signal of the investigated microorganism in the state under study. Along with a discussion of the possibilities of Raman spectroscopy for non-destructive analysis of microorganisms, ways are considered to increase the sensitivity of Raman spectroscopy in these studies.
