An ex vivo study of the kinetics of the optical properties of ovarian tissues under the influence of glycerol

Abstract
The optical properties of ovarian tissues of cats and dogs under the influence of glycerol were studied using diffuse transmission and reflection spectroscopy in a wide spectral range from UV to NIR. Immersion optical clearing during the impregnation of biological tissues with such a hyperosmotic agent as glycerol has a great effect on the optical properties of tissues. The principle of diffuse spectroscopy is based on the ability of tissue biological molecular chromophores (hemoglobin, oxyhemoglobin, porphyrins, collagen, DNA, etc.) to absorb diffusely scattered light of a certain wavelength. Scattering is a key characteristic of the transport and attenuation of light in tissues, especially in UV. Based on experimental data, using the free diffusion model and the modified Bouguer-Lambert-Beer law, the diffusion coefficient of glycerol into ovarian tissue ex vivo was calculated. The kinetics of optical clearing of a biological tissue was studied by recording the total transmission coefficients in the range from 200 to 800 nm. By combining the immersion method with UV spectroscopy, it was possible to test and study the main mechanisms of optical clearing - tissue dehydration and refractive index matching, and also found that the efficiency of optical clearing is much higher in deep UV than in the visible range. These studies can be used in the development of clinical methods for diagnosing the pathology of biological tissue at the cellular level.
