Laser Doppler flowmetry in assessing the effect of liraglutide on microcirculation in rats with diet-induced obesity

Abstract

Liraglutide (Lg) is a glucagon-like peptide-1 (GLP-1) analogue. GLP-1 inhibits gastric motility and emptying, resulting in reduced food intake and weight loss. The positive effect of Lg on microcirculation (MC) in diabetes mellitus has also been shown. Тhe pupose of the study was to investigate the anti-obesity effect of liraglutide in cafeteria diet-induced obesity, as well as its effect on microcirculation in obese white outbred rats.
The studies were performed on 30 white rats, divided into groups: 1) control (intact animals); 2) comparison group - animals with the diet-induced obesity (cafeteria diet); 3) experimental group - animals with the obesity, which were injected by Lg at dosage of 0.4 mg/kg/day. The anti-obesity effect of Lg, as well as its effect on МС in obese rats, was evaluated using calculation of the Lee index, increase in body weight, target organs, visceral fat, using the method of laser Doppler fluometry on the skin of the paw, the perfusion coefficient (М) was measured. Wavelet analysis was used to determine the amplitudes normalized by standard deviation of endothelial, neurogenic, myogenic, respiratory, pulse oscillations.
Lg reduced the Lee index, body weight relative to the rats of the comparison group. It was also found that Lg increased M, as well as the amplitudes of neurogenic and endothelial oscillations, and thus normalized perfusion of the skin of the paws of rats.
It has been shown that Lg is a potential clinical agent for the treatment of obesity and its microcirculatory disorders.
