Optimization of Airy beam parameters to improve light-sheet microscopy illumination

Abstract

Light-sheet microscopy (LSM) is a technique for 3D imaging of bio-samples and has facilities of fast, high contrast and minimal light exposure. Conventional Gaussian beam illumination has a rapid divergence so it prevents a long range uniform light-sheet. Beam shaping is widely noticeable for modification of the beam properties. Airy beam shows up increase in FOV because it remains diffraction-free over long distances compared to the Gaussian beam. Besides, the self-healing property minimizes the strip artifact generation. Combining the Airy beam with light sheet microscopy could potentially improve field of view and light penetration depth. In this research we discuss the effect of Airy beam characteristics generated by spatial light modulator (SLM) on the sheet properties of Airy-light-sheet microscopy (ALSM). Beam characteristics are controlled by computer generated hologram (CGH) patterns.
