Detection of spheroids formed in microfluidic wells using acoustic resolution photoacoustic microscopy

Abstract

Recent developments in tumor cells have heightened the need for 3D viewing, label-free, and depth imaging of spheroids. Photoacoustic microscopy (PAM) is an important imaging method and plays a key role in 3D depth imaging. In this study, we used acoustic resolution PAM to form 3D viewing of spheroids in a microfluidic channel. Likewise, the throughputs, size uniformity, easy handling, and long-term perfusion of spheroids are important so that we use microfluidic chips to form the uniform spheroids. The MCF7 tumor cells were injected and incubated into microfluidic chips. After aggregation of the cells, the spheroids were formed and the morphology of them was captured using an AR-PAM setup.
